Brief Description:

RG2 cells were grown to a sub-confluent monolayer. First, an aliquot of the primary library in washing/blocking buffer was added to an empty flask (depletion flask) to remove plastic-binding phage clones. Before incubation with RG2 cells, preselection steps with fibroblasts, myoblasts, and hepatocytes were included in the protocol to remove phage binding to these normal cells. Phage associated with RG2 cells were recovered from a single flask with elution buffer.
