Two phage libraries were then constructed (Creative Biolabs, Shirley, NY) in which TMT-2 (TLSPKMPGGGYW) formed the backbone on the M13KE RF vector. To the N-terminus of each bacteriophage 12-mer backbone, six amino acids were added at random using the NNK synthesis strategy, resulting in 1.0e8 distinct 18-mers in each library.
