TUPDB is a comprehensive database of target-unrelated peptides (TUPs) and TUP motifs. It contains extensive information about 2036 TUP sequences/motifs manually extracted from research articles and public databases.

Introduction


Affinity selection by phage display biopanning is successful in isolating target-specific ligands. However, the emergence of target-unrelated peptides (TUPs) has been hampering library screening. Those TUPs are false positive results. Besides target-specific binders, peptides binding to other constituents of the biopanning system other than the target molecule may also be recovered during the selection stage. These peptides are refereed as to selection-related TUPs (SrTUPs), which may dominant the screening results and prevent target binders identified. Multiple researchers have summarized and reported this type of TUPs. Another common type of TUPs have been characterized by faster propagation rates and are known as propagation-related TUPs (PrTUPs). The replication advantage of phage allows them to be favored during the amplification step. Isolation of such clones has no relation to their affinity.

TUP diagnosis is as important as the target-binding peptide discovery. Detailed review of the literature has revealed multiple TUPs. The biopanning community desperately needs a repository for harboring information related to experimentally-validated and suspected TUP sequences. Such a database will be important for TUP analysis and data mining.
