Assay Description: | To detect the inhibitory activity of peptides on infection of pseudotyped SARS-CoV-2, target cells (ACE2–293T) were plated at a density of 10^4 cells/well in a 96-well plate 12 h before infection. Diluted pseudotyped SARS-CoV-2 was mixed with peptides that were series diluted with DMEM without FBS and incubated at 37 °C for 30 min. The mixture was transferred to the ACE2-293T cells and incubated for 12 h. After that, the supernatant was replaced with complete medium and incubation continued for 48 h. Luciferase activity was analyzed by using the Luciferase Assay System (Promega), which refers to the amount of pseudotyped SARS-CoV-2 infecting target cells. |
